Oh MY... Quick vacation and NYC don't exactly mesh... There is SO MUCH to see and do, one could spend a month there and only have a good start.
My wife and I were married in NYC 20 yrs ago... so we have a special affinity for the place. She also has a brother and sister that live nearby and work in the city, so we go back often. In fact, she was just there two weeks ago and we go back in August for 5 days to visit and partake in the culture.
Some of the highlights that we enjoy:
The Palm Restaurant - no better steak or experience... The waiters (all men) will take an order of a table of 20 with no note pad, and place every item right where it is supposed to be... There are 2 Palms in NYC... (we had our wedding night dinner at PalmTwo; superb).

Best Dive in NYC: Lucy's Bar- Like you are stepping back into 1970 or so... and the food and drinks are cheap as 1970 too. Great burgers, huge sandwiches, fries, etc... juke box... you would never know you are in Manhatten...

Strolling Central Park, visit the site of WTC (somber), shopping 5th Ave, Times Square people watching, street vendors, rise early and visit the Today Show at Rockefeller Ctr, tour the NBC Studios there too (enlightening), China Town, Soho District, Empire State Bldg, Garnd Central Station (there is also a great casual restaurant right under the roadway @ GCS, excellent for breakfast or lunch; Grand Central Cafe, I think it is called).
